CARPE DIEM: NEW METHODOLOGIES FOR MONITORING MARMOSET NEUROENDOCRINE AND METABOLIC FUNCTION

     The common marmoset, Callithrix jacchus, has distinct advantages as a research primate for biomedical, behavioral and neuroendocrine studies. Marmosets are adult by 2 years of age, have a high rate of fertility and a short lifespan (~16 years). Additionally, twin births are common allowing for built in controls. Unfortunately, due to their small size, these small monkeys have limited blood volume to use for measurement of hormones and biomarkers. The Assay Services Laboratory at the Wisconsin National Primate Research Center has developed many new hormonal and biomarker assays that provide for multiple analyses on small volumes of blood. Use of the common marmoset for studies of obesity and metabolic syndrome requires markers of metabolic function. We have adapted commercial assays using small volumes of serum for measuring insulin, adiponectin , leptin and ghrelin requiring less than 250 µl for all measurements. These assays are specific and sensitive for marmoset samples. We have additionally developed new methodologies for measuring the urinary cortisol/cortisone ratio for use in determining glucoregulatory functioning by use of LC/MS (liquid chromatography/mass spectrometry). For neuroendocrine analyses it is common to require multi-steroid analysis and our new LC/MS method allows for measuring 10 steroids simultaneously in less than 200 µl of serum/plasma (cortisol, cortisone, corticosterone, testosterone, androstenedione, DHT, DHEA, estradiol, estrone and progesterone) with great sensitivity.
